兔來源免疫組化用抗體保存條件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
兔來源免疫組化用抗體,Anti-GAPDH抗體抗原修復(fù)方法:
方法1:沸水浴修復(fù),將盛有修復(fù)液和玻片的燒杯置于沸水浴環(huán)境,保持外部沸騰狀態(tài)15min,自然冷卻至室溫。
方法2:微波修復(fù),將盛有修復(fù)液和玻片的燒杯置于微波爐中,高火5min,停火3min,中火5min,自然冷卻至室溫。
方法3:高壓修復(fù),修復(fù)液加入高壓鍋加熱至沸騰,放入玻片,封蓋加壓持續(xù)加熱至噴氣時開始計時修復(fù)2min,自然冷卻至室溫。
方法4:使用*修復(fù),將*修復(fù)液加入到組織上,37℃,消化30min

抗原修復(fù),必須注意以下問題:
1、修復(fù)液的選擇。
0.01M枸櫞酸(pH6.0):應(yīng)用zui多的抗原修復(fù)液,適用于大多數(shù)的抗原,用該液修復(fù)后的抗原表達增強,抗原的定性和定位很理想,結(jié)果不錯;
0.05M EDTA(pH8.0):應(yīng)用較多的抗原修復(fù)液,修復(fù)能力較強,對于保存時間較長的切片或目的蛋白表達較弱的組織有很好的修復(fù)效果,需要控制好修復(fù)時間,否則容易出現(xiàn)較重的背景;
0.01M TBS(pH7.4):中性修復(fù)液,適用于大多數(shù)抗原,對于核定位蛋白有較好的修復(fù)效果;
*:應(yīng)用較多的抗原修復(fù)液,適用于大多數(shù)的抗原,使用溫度37℃,對于容易脫片的切片有很好的保護作用。

2.抗原熱修復(fù)時應(yīng)選擇的溫度。
據(jù)實驗認為溫度在92℃-98℃是合適的,尤其在95℃為,這是因為:(1)這種溫度未達沸騰,切片不容易脫離載玻片;(2)能夠解離和破壞與蛋白交聯(lián)的甲醛醛鍵等,處理時可以隨意選擇和確定抗原修復(fù)的作用時間。如果選擇高壓修復(fù),因為溫度較高,時間不宜過長
3.抗原修復(fù)時有效溫度所需持續(xù)時間根據(jù)各單位的設(shè)備條件以及使用的儀器不同,抗原修復(fù)時在有效的溫度范圍內(nèi)所持續(xù)的時間也不一樣。
4.抗原修復(fù)液必須遵循自然降溫規(guī)律,否則效果不好或達不到抗原修復(fù)的目的。
5.盡量使用足量的抗原修復(fù)液,防止切片干涸。
6.切片必須附貼牢固,否則發(fā)生掉片。

背景介紹:Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is well known as one of the key enzymes involved in glycolysis. It catalyzes an important energy-yielding step in carbohydrate metabolism, thereversible oxidative phosphorylation of glyceraldehyde-3-phosphatein the presence of inorganic phosphate and nicotinamide adenine dinucleotide (NAD). Recent evidence suggests that it also is involved in a number of cellular processes such as membrane fusion, phosphotransferase activity, DNA replication and repair, and nuclear RNA export (1). GAPDH has also been implicated in playing a role in different pathologies such as cancer progression, apoptosis, and neuronal diseases such as Alzheimer’s and Huntington’s disease (2). GAPDH is constitutively expressed at high levels in almost all tissues and cell lines making it ideal for use as a loading control marker in immunoblots.
