瘦素受體抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

石蠟切片的免疫組化SP法參考步驟：
1.60℃烤片2小時
2.脫蠟。二甲苯脫蠟15分鐘，三次
3.水化。依次經過無水乙醇、95%、90%、80%、70%，各5分鐘，蒸餾水（或自來水）5分鐘
4.PBS洗5分鐘，三次
5.抗原修復。枸櫞酸緩沖液（約92-95℃）煮沸（沸水浴）修復15分鐘，自然涼至室溫。PBS洗5分鐘，三次
6.3%雙氧水消化內源性的過氧化物酶，20分鐘，PBS洗5分鐘，三次
7.正常山羊血清封閉，37℃20分鐘
8.一抗孵育，4℃過ye.
9.PBS洗5分鐘，三次
10.二抗孵育，37℃20分鐘（二抗為*標記的羊抗兔IgG），PBS洗5分鐘，三次
11.三抗孵育，37℃20分鐘（三抗為辣根酶標記的鏈霉親和素），PBS洗5分鐘，三次
12.DAB顯色，鏡下觀察結果，適時終止
13.蘇木素復染5分鐘，自來水沖洗片刻，1%的鹽酸酒精（75%）溶液（請自行配置，配制比例為：100ml 75%酒精+1ml濃鹽酸）分化30秒，自來水沖洗5分鐘藍化
14.脫水。依次經過70%、80%、90%、95%、無水乙醇，各5分鐘，二甲苯15分鐘，三次
15.封片，中性樹膠封片

瘦素受體抗體,Anti-Leptin receptor抗體溶解方法:

方法1：我們的抗體（凍干粉）已經包含了0.01M PBS、1% BSA和0.1% 防腐劑（疊氮鈉或*），您只需要加入滅菌的雙蒸水就行了。抗體溶解后，置于-20℃保存，分裝后使用，避免反復凍融，可保證至少1年有效；

方法2：也可以只加入一半體積的雙蒸水，再用一半體積的甘油補足，置于-20℃保存，這樣抗體不會凍，有利于抗體的穩定。

后續試驗時，再使用對應的緩沖液稀釋成工作液，即用即配。

我們的抗體（凍干粉），在常溫放置下，至少一個月有效；若在-20℃下保存（*），至少三年有效。

Antibodies are prepared with 0.01M PBS, pH 7.4 with 1% BSA and 0.1% Sodium Azide, prior to lyophilization. Currently, we offer both lyophilized and reconstituted antibodies, both resulting in final concentration of 1mg/mL. We recommend two ways to reconstitute antibodies in lyophilized form:

1) Reconstituting by directly adding 100uL of sterile, distilled water.

2) Alternatively, if glycerol does not affect the experiment, add 50uL sterile distilled water, followed by adding 50uL of glycerol.

背景介紹：The protein encoded by this gene belongs to the gp130 family of cytokine receptors that are known to stimulate gene transcription via activation of cytosolic STAT proteins. This protein is a receptor for leptin (an adipocyte-specific hormone that regulates body weight), and is involved in the regulation of fat metabolism, as well as in a novel hematopoietic pathway that is required for normal lymphopoiesis. Mutations in this gene have been associated with obesity and pituitary dysfunction. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. It is noteworthy that this gene and LEPROT gene (GeneID:54741) share the same promoter and the first 2 exons, however, encode distinct proteins (PMID:9207021).
