Anti-AOA1抗體,共濟失調性眼球運動功能喪失相關蛋白AOA1抗體產品介紹：Aprataxin is a nuclear protein, present in both the nucleoplasm and the nucleolus, which is a member of the histidine triad (HIT) superfamily. Aprataxin is involved in DNA single-strand break repair, mediating protein-protein interactions with molecules responding to DNA damage. Aprataxin contains three conserved domains: an N-terminal forkhead-associated (FHA) domain which mediates protein-protein interactions, a HIT domain that is similar to Hint, and a C-terminal zinc finger domain. Loss of function mutations in APTX, the gene encoding for Aprataxin, destabilize the Aprataxin protein and result in a rare neurological disorder known as ataxia-oculomotor apraxia, characterized by abnormal movements of the head and eyes. These mutations either target the HIT domain or truncate the protein N-terminal to a zinc finger.

設置對照
其目的在于證明和肯定陽性結果的特異性，排除非特異性疑問。主要是針對*抗體對照，常用的對照方法包括：1. 陽性對照；2. 陰性對照；3. 阻斷實驗； 4.替代對照；5. 空白對照；6. 自身對照；7. 吸收實驗.
1. 陽性對照用已知抗原陽性的切片作對照與待檢標本同時進行免疫組織化學染色，對照切片應呈陽性結果，成為陽性對照。證明全過程均符合要求，尤其當待檢標本呈現陰性結果時，陽性對照尤為重要。
2.陰性對照用確證不含已知抗原的標本作對照，應呈陰性結果，成為陰性對照，是陰性對照的一種。其實空白、替代、吸收和抑制都屬于陰性對照。當待檢標本呈陽性結果時，陰性對照就更加重要，用以排除假陽性。
顯色觀察
免疫酶染色應注意控制：
1. 呈色質濃度和反應時間。增加成色質的量和（或）增加底物反應時間，可增加反應產物強度；著色太深可減少反應時間。
2. 過氧化物酶顯色時，H2O2濃度較大將使顯色反應過快而至背景加深；過量H2O2可能抑制酶的活性。
