Anti-BRCA1抗體品介紹：This gene encodes a nuclear phosphoprotein that plays a role in maintaining genomic stability, and it also acts as a tumor suppressor. The encoded protein combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as the BRCA1-associated genome surveillance complex (BASC). This gene product associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complexes. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in this gene are responsible for approximay 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Alternative splicing plays a role in modulating the subcellular localization and physiological function of this gene. Many alternatively spliced transcript variants, some of which are disease-associated mutations, have been described for this gene, but the full-length natures of only some of these variants has been described. A related pseudogene, which is also located on chromosome 17, has been identified.
免疫組化結果的判斷
對免疫組化結果的判斷應持科學的慎重態度，要準確判斷陽性和陰性，排除假陽性和假陰性結果，必須嚴格對照實驗，對新發現的陽性結果，除有對照實驗結果之外，應進行多次重復實驗，可用幾種方法進行驗證。必須學會判斷特異性染色和非特異性染色，對初學者更為重要，否則會得出不科學的結論。特異性染色和非特異性染色的鑒別點主要在于特異性反應產物常分布于特定的部位。如胞漿內，也有分布在細胞核和細胞表面的，即具有結構性。特異性染色表現為在同一切片上呈現不同程度的陽性染色結果。非特異性染色表現為無一定的分布規律，常呈某一部位成片的均勻著色，細胞和周圍的結締組織均無區別的著色，或結締組織呈現很強的染色。非特異性染色常出現在干燥切片的邊緣，有刀痕或者折疊的部位。在過大的組織塊，中心固定不良也會導致非特異性染色。有時可見非特異性染色和特異性染色同時存在，由于過強的非特異性染色背景不但影響對特異性染色結果的觀察和記錄，而且令人對其特異性結果產生懷疑。
