保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

Anti-Androgen receptor抗體,雄激素受體抗體產(chǎn)品介紹：The androgen receptor gene is more than 90 kb long and codes for a protein that has 3 major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. The protein functions as a steroid-hormone activated transcription factor. Upon binding the hormone ligand, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and then stimulates transcription of androgen responsive genes. This gene contains 2 polymorphic trinucleotide repeat segments that encode polyglutamine and polyglycine tracts in the N-terminal transactivation domain of its protein. Expansion of the polyglutamine tract causes spinal bulbar muscular atrophy (Kennedy disease). Mutations in this gene are also associated with complete androgen insensitivity (CAIS). Two alternatively spliced variants encoding distinct isoforms have been described.

失敗的幾種原因

1.所染的全部切片均為陰性結(jié)果，包括陽性對照在內(nèi)，全部呈陰性反應(yīng)，原因可能是：（1）染色未按嚴(yán)格操作步驟進行；（2）漏加一種抗體，或抗體失效；（3）緩沖液內(nèi)含*，抑制了酶的活性；（4）底物中所加H2O2量少或失效；復(fù)染或脫水劑使用不當(dāng)。
2.所有切片均呈弱陽性反應(yīng)：（1）切片在染色過程中抗體過濃，或干燥了；（2）緩沖液配制中未加氯化鈉或pH不準(zhǔn)確，洗滌不*；（3）使用已變色的呈色底物溶液，或呈色反應(yīng)時間過長；（4）抗體溫育的時間過長；（5）H2O2濃度過高，呈色速度過快；（6）粘附劑太厚。
3.所有切片背景過深：（1）未用酶消化處理切片；（2）切片或涂片過厚；（3）漂洗不夠；（4）底物呈色反應(yīng)過久；（5）蛋白質(zhì)封閉不夠或所用血清溶血；（6）使用全血清抗體稀釋不夠。
