Anti-AADAC抗體,芳香乙酰胺脫乙酰基酶抗體產(chǎn)品介紹：The assembly of very-low-density lipoproteins (VLDLs) in the secretory apparatus of the hepatocyte relies on the mobilization of triacylglycerol (TAG) from the cytosolic pool by lipolysis and re-esterification. However, some of the re-esterified TAG products are returned to the cytosolic pool in the liver, which protects vulnerable body tissues from excess lipotoxic non-esterified fatty acids in the plasma. Some of the lipases involved in this process include arylacetamide deacetylase (AADAC) and its related proteins AADACL1 and AADACL2. AADAC, a single pass type II membrane protein of the endoplasmic reticulum, is expressed in hepatocytes, intestinal mucosal cells, pancreas and adrenal gland. It plays an important role in the metabolic activation of arylamine substrates to ultimate carcinogens. AADACL1 hydrolyzes the metabolic intermediate 2-acetyl monoalkylglycerol, and its inactivation results in disruption of ether lipid metabolism in cancer cells and impaired cell migration and tumor growth.

對(duì)免疫組化結(jié)果的判斷應(yīng)持科學(xué)的慎重態(tài)度，要準(zhǔn)確判斷陽(yáng)性和陰性，排除假陽(yáng)性和假陰性結(jié)果，必須嚴(yán)格對(duì)照實(shí)驗(yàn)，對(duì)新發(fā)現(xiàn)的陽(yáng)性結(jié)果，除有對(duì)照實(shí)驗(yàn)結(jié)果之外，應(yīng)進(jìn)行多次重復(fù)實(shí)驗(yàn)，可用幾種方法進(jìn)行驗(yàn)證。必須學(xué)會(huì)判斷特異性染色和非特異性染色，對(duì)初學(xué)者更為重要，否則會(huì)得出不科學(xué)的結(jié)論。特異性染色和非特異性染色的鑒別點(diǎn)主要在于特異性反應(yīng)產(chǎn)物常分布于特定的部位。如胞漿內(nèi)，也有分布在細(xì)胞核和細(xì)胞表面的，即具有結(jié)構(gòu)性。特異性染色表現(xiàn)為在同一切片上呈現(xiàn)不同程度的陽(yáng)性染色結(jié)果。非特異性染色表現(xiàn)為無(wú)一定的分布規(guī)律，常呈某一部位成片的均勻著色，細(xì)胞和周?chē)慕Y(jié)締組織均無(wú)區(qū)別的著色，或結(jié)締組織呈現(xiàn)很強(qiáng)的染色。非特異性染色常出現(xiàn)在干燥切片的邊緣，有刀痕或者折疊的部位。在過(guò)大的組織塊，中心固定不良也會(huì)導(dǎo)致非特異性染色。有時(shí)可見(jiàn)非特異性染色和特異性染色同時(shí)存在，由于過(guò)強(qiáng)的非特異性染色背景不但影響對(duì)特異性染色結(jié)果的觀察和記錄，而且令人對(duì)其特異性結(jié)果產(chǎn)生懷疑。
