Anti-Ankyrin G 抗體,錨定蛋白G抗體產(chǎn)品介紹：human: Ankyrins are a family of proteins that are believed to link the integral membrane proteins to the underlying spectrin-actin cytoskeleton and play key roles in activities such as cell motility, activation, proliferation, contact, and the maintenance of specialized membrane domains. Multiple isoforms of ankyrin with different affinities for various target proteins are expressed in a tissue-specific, developmentally regulated manner. Most ankyrins are typically composed of three structural domains: an amino-terminal domain containing multiple ankyrin repeats; a central region with a highly conserved spectrin binding domain; and a carboxy-terminal regulatory domain which is the least conserved and subject to variation. Ankyrin 3 is an immunologically distinct gene product from ankyrins 1 and 2, and was originally found at the axonal initial segment and nodes of Ranvier of neurons in the central and peripheral nervous systems. Alternatively spliced variants may be expressed in other tissues. Although multiple transcript variants encoding several different isoforms have been found for this gene, the full-length nature of only two have been characterized. [provided by RefSeq]
mouse: This gene encodes a member of the ankyrin protein family. Ankyrins link integral membrane proteins to the spectrin-based Ankyrins link integral membrane proteins to the spectrin-based cytoskeleton. Ankyrin family members share a protein structure which includes three independently folded domains: the N-terminal ankyrin repeat domain, the central spectrin-binding domain, and the C-terminal rod domain. This ankyrin functions as the major ankyrin in the kidney and may play a role in the polarized distribution of many integral membrane proteins to specific subcellular sites. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms.

合適的抗體稀釋度：

抗體的濃度是免疫染色的關鍵，如果抗體濃度過高，抗體分子過多于抗原決定簇，可導致抗體結(jié)合減少，產(chǎn)生陰性結(jié)果。此陰性結(jié)果并不一定是缺少抗原，而是由于抗體過量，這種現(xiàn)象類似于凝集反應中的前帶效應（prozone effect）。因此，必須使用一系列稀釋做“棋盤式效價滴定”，檢測抗體的合適稀釋度，以得到zui大強度的特異性染色和zui弱的背景染色。抗體稀釋度應根據(jù)：1.抗體效價高，溶液中特異性抗體濃度越高，工作稀釋度越高；2.一般講，應用的抗體稀釋度越大，溫育時間越長；3.對于抗體與非特異性蛋白的結(jié)合，只有高稀釋度時才能防止其非特異性背景染色；4.稀釋用緩沖液的種類，標本的固定和處理過程等也可影響稀釋度，所以合適的稀釋度應根據(jù)具體情況測定。抗體的稀釋主要是指*抗體，因為*抗體中特異性抗體合適的濃度是關鍵，應用高稀釋度*抗體僅高親和力的特異性染色反應，減少或消除其中交叉抗體反應。

抗原修復方法：
方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫。
方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
方法3：高壓修復，修復液加入高壓鍋加熱至沸騰，放入玻片，封蓋加壓持續(xù)加熱至噴氣時開始計時修復2min，自然冷卻至室溫。
方法4：使用*修復，將*修復液加入到組織上，37℃，消化30min
