小鼠牛血清白蛋白（BSA）ELISA試劑盒實驗測定具體步驟：
1．確定本次檢測所需的已包被抗體的酶標板孔數目，并增加 1 孔作為 TMB 空白顯色孔。 總數=樣品數+9；做雙份檢測時×2。其余重包裝好放如冰箱中。 
2．將標準品各 0.1ml 依次加入一排 7 孔中，1 孔只加樣品稀釋液的作為零孔。對于血清、體液、 組織勻漿或細胞培養上清，直接加已用樣品稀釋液稀釋的樣品 100μ ι 。 
3．酶標板加上蓋，37℃反應 90 分鐘。 
4．反應后用自動洗板機吸去酶標板內的液體；或甩去酶標板內液體，再對著吸水紙拍幾下。 不洗。 
5．將準備好的生物素抗體工作液按每孔 0.1ml 依次加入。（TMB 空白顯色孔除 外）。37℃反應 60 分鐘。 
6． 0.01M TBS 或 0.01M PBS 洗滌 3 次，每次浸泡 1 分鐘左右。 
7．將準備好的 ABC 工作液按每孔 0.1ml 依次加入（TMB 空白顯色孔除外）。37℃反應 30 分鐘。 
8． 0.01M TBS 或 0.01M PBS 洗滌 5 次，每次浸泡 1-2 分鐘左右。 
9．按每孔 90μ ι 依次加入已在 37℃平衡 30 分鐘的 TMB 顯色液，37℃避光反應 20-25 分鐘 （注意：顯色時間供參考，因用戶實驗室條件差異，顯色時間會有所不同。此時肉眼可 見標準品的前 3-4 孔有明顯的梯度藍色，后 3-4 孔差別不明顯）。 
10．按每孔 0.1ml 依次加入 TMB 終止液，此時藍色立轉黃色。 
11．用酶標儀在 450nm 測定 OD值。

有兩種設定空白對照的方案： 
（1）將 TMB 空白顯色孔設為對照。所有的標準品和樣品的吸光值減去 TMB 空白顯色孔的 吸 光 值 后 ， 在 坐 標 紙 上 畫 出 曲 線 ， 以 吸 光 值 作 為 縱 坐 標 ， 以 濃 度 作 為 橫 坐 標 。 
（2）將零孔設為對照。所有的標準品和樣品的吸光值減去零孔的吸光值后，得到的數據可以 直接在坐標紙上畫出曲線。 

樣品稀釋的一般原則 
用戶須估計樣品待測因子的含量，決定適當的稀釋倍數，以使稀釋后樣品中待測因子的濃度 處于 ELISA 試劑盒的檢測范圍。根據待測因子含量高、中、低的不同，分別采取不同的稀釋方案：
高：指待測因子在 20-200ng/ml。一般按 1：100 稀釋。297μ ι 樣品稀釋液加 3μ ι 樣品。 
中：指待測因子在 2-20ng/ml。一般按 1：10 稀釋。225μ ι 樣品稀釋液加 25μ ι 樣品。 
低：指待測因子在 31.2→2000pg/ml。按 1：2 稀釋。100μ ι 樣品稀釋液加 100μ ι 樣品。 
特低：指待測因子≤31.2pg/ml。樣品一般不做稀釋，按每孔 100μ ι 樣品直接加入。

以上方案僅供參考，樣品的稀釋應有詳細記錄。
