影響ELISA中非特異性顯色的原因有很多，比如試劑盒特異性、檢驗(yàn)標(biāo)本中含酶標(biāo)記物的干擾物，操作過程中的問題。現(xiàn)在就這原因作一簡要分析,以便可以通過以上措施把非特異顯色降至zui低限度，從而提高檢測的特異性，并得到更準(zhǔn)確、可靠的實(shí)驗(yàn)結(jié)果。

elisa 小知識：
辣根過氧化物酶(HRP)是一種糖蛋白，每個(gè)分子含有一個(gè)氯化血紅素(protonhemin)區(qū)作輔基。酶的濃度和純度常以輔基的含量表示。氯化血紅素輔基的zui大吸收峰是403nm，HRP酶蛋白的zui大吸收峰是275nm，所以酶的濃度和純度計(jì)算式是(已知HRP的A(1cm 403nm 1%)＝25，式中1％指HRP百分濃度為100ml含酶蛋白1g，即10mg/ml，所以，酶濃度以 mg/ml 計(jì)算是HRP的A(1cm 403nm mg/ml=2.5)HRP純度(RZ)=A403nm/A275nm純度RZ(Ｒeinheit Zahl)值越大說明酶內(nèi)所含雜質(zhì)越少。高純度HRP的RZ值在3.0左右，zui高可達(dá)3.4。用于ELISA檢測的HRP的RZ值要求在3.0以上。
