ELISA實(shí)驗(yàn)操作注意事項(xiàng)：
大部分實(shí)驗(yàn)室在使用酶標(biāo)儀之前是通過目測(cè)判斷結(jié)果，操作過程隨意性較大，在使用酶標(biāo)儀后操作必須規(guī)范：
A. 使用移液器加液，移液槍頭不能混用。
B. 洗板要干凈，避免交叉污染。
C. 嚴(yán)格按照試劑盒的說明書操作，反應(yīng)時(shí)間準(zhǔn)確。
D. 請(qǐng)勿將樣品或試劑灑到儀器表面或內(nèi)部，操作完成注意做好清潔工作。
E. 不要在測(cè)量過程中關(guān)閉電源。
F. 對(duì)于因試劑盒問題造成的測(cè)量結(jié)果的偏差，應(yīng)根據(jù)實(shí)際情況及時(shí)修改參數(shù)，以達(dá)到*效果。
G. 使用后蓋好防塵罩。
H. 出現(xiàn)技術(shù)故障時(shí)應(yīng)及時(shí)與廠家，切勿擅自拆卸酶標(biāo)儀。

ELISA小常識(shí)：
間接法是檢測(cè)抗體zui常用的方法，其原理為利用酶標(biāo)記的抗抗體以檢測(cè)已與固相結(jié)合的受檢抗體，故稱為間接法。操作步驟如下：(1)將特異性抗原與固相載體連接，形成固相抗原(2)加稀釋的受檢血清：其中的特異抗體與抗原結(jié)合，形成固相抗原抗體復(fù)合物。(3)加酶標(biāo)抗抗體：與固相復(fù)合物中的抗體結(jié)合，從而使該抗體間接地標(biāo)記上酶。(4)加底物顯色：顏色深度代表標(biāo)本中受檢抗體的量。
