用途:CAS 59-05-2,氨甲蝶呤用于生化研究。*激動劑。依靠阻斷合成胸腺嘧啶核苷必須的四氫*輔助因子來阻斷DNA的合成。氨甲蝶呤靠*轉(zhuǎn)運體轉(zhuǎn)運進細胞。在細胞里,它被葉酰聚*合成酶(folylpolyglutamate synthase)轉(zhuǎn)化成高分子量的聚*代謝物,又結(jié)合到二氫*還原酶(dihydrofolate reductase)上,從而抑制了它的活性。氨甲喋呤-聚*被γ-谷氨酰水解酶(γ-glutamyl hydrolasen)在細胞內(nèi)降解。
