CREB結合蛋白p300抗體背景介紹:Cyclic AMP-regulated gene expression frequently involves a DNA element designated the cAMP-regulated enhancer (CRE). Many transcription factors bind to this element, including the protein CREB which is activated as a result of phosphorylation by protein kinase A. It has been shown that protein kinase A-mediated CREB phosphorylation results in its binding to a nuclear protein designated CBP (for CREB-binding protein). These findings suggest that CBP has many of the properties expected of a CREB co-activator. Another high molecular weight transcriptional adapter protein, designated p300, is characterized by three cysteine- and histidine-rich regions, of which the most carboxy terminal region specifically binds the adenovirus E1A protein. p300 molecules lacking an intact E1A binding site bypass E1A repression even in the presence of high concentrations of E1A. Sequence analysis of CBP and p300 has revealed substantial homology, arguing that these proteins are members of a conserved family of co-activators.

抗體修復方式
修復液:0.01M 枸櫞酸(pH6.0)或者0.05M EDTA(pH8.0)
CREB結合蛋白p300抗體,Anti-Ep300抗體抗原修復,必須注意以下問題:
1、修復液的選擇。
0.01M枸櫞酸(pH6.0):應用zui多的抗原修復液,適用于大多數的抗原,用該液修復后的抗原表達增強,抗原的定性和定位很理想,結果不錯;
0.05M EDTA(pH8.0):應用較多的抗原修復液,修復能力較強,對于保存時間較長的切片或目的蛋白表達較弱的組織有很好的修復效果,需要控制好修復時間,否則容易出現較重的背景;
0.01M TBS(pH7.4):中性修復液,適用于大多數抗原,對于核定位蛋白有較好的修復效果;
*:應用較多的抗原修復液,適用于大多數的抗原,使用溫度37℃,對于容易脫片的切片有很好的保護作用。

2.抗原熱修復時應選擇的溫度。
據實驗認為溫度在92℃-98℃是合適的,尤其在95℃為,這是因為:(1)這種溫度未達沸騰,切片不容易脫離載玻片;(2)能夠解離和破壞與蛋白交聯的甲醛醛鍵等,處理時可以隨意選擇和確定抗原修復的作用時間。如果選擇高壓修復,因為溫度較高,時間不宜過長
3.抗原修復時有效溫度所需持續時間根據各單位的設備條件以及使用的儀器不同,抗原修復時在有效的溫度范圍內所持續的時間也不一樣。
4.抗原修復液必須遵循自然降溫規律,否則效果不好或達不到抗原修復的目的。
5.盡量使用足量的抗原修復液,防止切片干涸。
6.切片必須附貼牢固,否則發生掉片。
