保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

Anti-MTMR14抗體,肌管素相關蛋白14抗體產(chǎn)品介紹：Myotubularin-related protein 14 (MTMR14), also known as Jumpy, is a myotubularin-related phosphoinositol-3-phosphate (PI3P) phosphatase (1). Mutations in the MTMR14 gene have been associated with centronuclear myopathy (1). MTMR14 deficiency in mice leads to altered calcium homeostasis and muscle disorders (2). MTMR14 has also been shown to play a role in autophagy, a process that is highly regulated by phosphatidylinositides through the type III PI3K, Vps34 (3). MTMR14 was localized to autophagic isolation membranes and early autophagosomes (3). In these studies, MTMR14 inhibited autophagy and mutations of MTMR14 associated with centronuclear myopathy were also defective in autophagy inhibition. In zebrafish, MTMR14 knockdown was shown to increase the number of autophagosomes, suggesting that its activity is associated with an inhibition of autophagy (4).

抗原修復，必須注意以下問題：

1、修復液的選擇。
0.01M枸櫞酸（pH6.0）：應用zui多的抗原修復液，適用于大多數(shù)的抗原，用該液修復后的抗原表達增強，抗原的定性和定位很理想，結果不錯;
0.05M EDTA（pH8.0）：應用較多的抗原修復液，修復能力較強，對于保存時間較長的切片或目的蛋白表達較弱的組織有很好的修復效果，需要控制好修復時間，否則容易出現(xiàn)較重的背景;
0.01M TBS（pH7.4）：中性修復液，適用于大多數(shù)抗原，對于核定位蛋白有較好的修復效果;
*：應用較多的抗原修復液，適用于大多數(shù)的抗原，使用溫度37℃，對于容易脫片的切片有很好的保護作用。

2.抗原熱修復時應選擇的溫度。
據(jù)實驗認為溫度在92℃－98℃是合適的，尤其在95℃為，這是因為：（1）這種溫度未達沸騰，切片不容易脫離載玻片；（2）能夠解離和破壞與蛋白交聯(lián)的甲醛醛鍵等，處理時可以隨意選擇和確定抗原修復的作用時間。如果選擇高壓修復，因為溫度較高，時間不宜過長

3.抗原修復時有效溫度所需持續(xù)時間根據(jù)各單位的設備條件以及使用的儀器不同，抗原修復時在有效的溫度范圍內(nèi)所持續(xù)的時間也不一樣。
4.抗原修復液必須遵循自然降溫規(guī)律，否則效果不好或達不到抗原修復的目的。
5.盡量使用足量的抗原修復液，防止切片干涸。
6.切片必須附貼牢固，否則發(fā)生掉片。
