英文介紹：This gene encodes a member of the fascin family of actin-binding proteins. Fascin proteins organize F-actin into parallel bundles, and are required for the formation of actin-based cellular protrusions. The encoded protein plays a critical role in cell migration, motility, adhesion and cellular interactions. Expression of this gene is known to be regulated by several microRNAs, and overexpression of this gene may play a role in the metastasis of multiple types of cancer by increasing cell motility. Expression of this gene is also a marker for Reed-Sternberg cells in Hodgkin's lymphoma. A pseudogene of this gene is located on the long arm of chromosome 15.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

Anti-Fascin1抗體,纖維束1抗體免疫組化染色：

封閉(Blocking)

轉(zhuǎn)膜完畢后，立即把蛋白膜放置到預(yù)先準(zhǔn)備好的Western洗滌液中，漂洗1-2分鐘，以洗去膜上的轉(zhuǎn)膜液。從轉(zhuǎn)膜完畢后所有的步驟，一定要注意膜的保濕，避免膜的干燥，否則極易產(chǎn)生較高的背景。
用微型臺(tái)式真空泵吸盡洗滌液，加入Western封閉液，在搖床上緩慢搖動(dòng),室溫封閉60分鐘。對(duì)于一些背景較高的抗體，可以在4℃ 封閉過(guò)夜。

一抗孵育(Primary antibody incubation)

參考一抗的說(shuō)明書(shū)，按照適當(dāng)比例用Western一抗稀釋液稀釋一抗。
用微型臺(tái)式真空泵吸盡封閉液，立即加入稀釋好的一抗，室溫或4℃在側(cè)擺搖床上緩慢搖動(dòng)孵育一小時(shí)。如果一抗孵育一小時(shí)效果不佳，可以在4℃緩慢搖動(dòng)孵育過(guò)夜。回收一抗。加入Western洗滌液，在側(cè)擺搖床上緩慢搖動(dòng)洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長(zhǎng)洗滌時(shí)間并增加洗滌次數(shù)。

二抗孵育(Secondary antibody inucubation)

參考二抗的說(shuō)明書(shū)，按照適當(dāng)比例用Western二抗稀釋液稀釋辣根過(guò)氧化物酶(HRP)標(biāo)記的二抗。
用微型臺(tái)式真空泵吸盡洗滌液，立即加入稀釋好的二抗，室溫或4℃在側(cè)擺搖床上緩慢搖動(dòng)孵育一小時(shí)。回收二抗，加入Western洗滌液，在側(cè)擺搖床上緩慢搖動(dòng)洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長(zhǎng)洗滌時(shí)間并增加洗滌次數(shù)。

蛋白檢測(cè)(Detection of proteins)

參考相關(guān)說(shuō)明書(shū)，使用BeyoECL, Western 熒光檢測(cè)試劑等ECL類(lèi)試劑來(lái)檢測(cè)蛋白。
洗片時(shí)可以使用X光片自動(dòng)洗片機(jī)。如果沒(méi)有自動(dòng)洗片機(jī)，可以用顯影定影試劑盒自行配制顯影液和定影液進(jìn)行手工洗片。

膜的重復(fù)利用(Membrane recovery)

如果蛋白樣品非常寶貴，可以使用Western一抗二抗去除液處理蛋白膜，以重復(fù)利用蛋白膜。
