英文介紹：The protein encoded by this gene is a pleiotropic cytokine that functions as a chemoattractant, a modulator of T cell activation, and an inhibitor of HIV replication. The signaling process of this cytokine is mediated by CD4. The product of this gene undergoes proteolytic processing, which is found to yield two functional proteins. The cytokine function is exclusively attributed to the secreted C-terminal peptide, while the N-terminal product may play a role in cell cycle control. Caspase 3 is reported to be involved in the proteolytic processing of this protein. Alternate splicing results in multiple transcript variants.

保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

一抗孵育(Primary antibody incubation)
參考一抗的說明書，按照適當(dāng)比例用Western一抗稀釋液稀釋一抗。
用微型臺式真空泵吸盡封閉液，立即加入稀釋好的一抗，室溫或4℃在側(cè)擺搖床上緩慢搖動孵育一小時。如果一抗孵育一小時效果不佳，可以在4℃緩慢搖動孵育過夜?；厥找豢?。加入Western洗滌液，在側(cè)擺搖床上緩慢搖動洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長洗滌時間并增加洗滌次數(shù)。
二抗孵育(Secondary antibody inucubation)
參考二抗的說明書，按照適當(dāng)比例用Western二抗稀釋液稀釋辣根過氧化物酶(HRP)標(biāo)記的二抗。
用微型臺式真空泵吸盡洗滌液，立即加入稀釋好的二抗，室溫或4℃在側(cè)擺搖床上緩慢搖動孵育一小時?；厥斩?，加入Western洗滌液，在側(cè)擺搖床上緩慢搖動洗滌5-10分鐘。吸盡洗滌液后，再加入洗滌液，洗滌5-10分鐘。共洗滌3次。如果結(jié)果背景較高可以適當(dāng)延長洗滌時間并增加洗滌次數(shù)。
蛋白檢測(Detection of proteins)
參考相關(guān)說明書，使用BeyoECL, Western 熒光檢測試劑等ECL類試劑來檢測蛋白。
洗片時可以使用X光片自動洗片機。如果沒有自動洗片機，可以用顯影定影試劑盒自行配制顯影液和定影液進行手工洗片。
